This review explains that for people with genetic forms of obesity, including some individuals with 16p11.2 deletion–related obesity, medications that target appetite biology (such as GLP‑1 medicines and, in specific genetic cases, pathway‑targeted drugs) can meaningfully reduce hunger and weight when lifestyle changes alone are not enough—helping families know that medical treatment can be an appropriate, biology‑based support to discuss with an endocrinologist, not a failure of effort or parenting.
