Integrating de novo and inherited variants in 42,607 autism cases identifies mutations in new moderate-risk genes.

This very large genetics study shows that autism—including in children with 16p11.2 syndromes—often reflects the combined effect of a major genetic change plus many additional inherited risk genes, helping families understand why abilities and challenges vary so much and why care plans should be personalized rather than based on a single genetic finding alone.
